Duplicate Card Detection
I use multiple sources for my card data, and sometimes the exact same question will appear in more than one source. I can end up with duplicate cards and find that it would be help ful to be able to perform duplicate card detection once cards have been entered. This will also be a useful function once the ability to export/import cards has also been implemented.

Universal off-line mobile client (500 work units)
Use javascript to write a mobile client which should work on Android, iPhone and Windows Phone 7.
How well this cross-platform aspect works in practice still remains to be seen, but it's the only option given the size of the core development team.
Initially, it will be a thin client very much like MnemoDodo, where you only export a few days' worth of revisions. Difference with MnemoDodo will be that the sync can be initiated from the phone

UI support for N-sided card types (30 work units)
Under the hood, libmnemosyne already supports this, but currently, there is no UI support.
This will enable card types with a arbitrary number of fields, and with an aribtrary number of sister cards.
Support will also be added for custum CSS and custom standard text on each of the cards.

Edit tags
Provide some way to edit existing tags.
Currently tags can only added or removed from cards. What if the tag has a typo or you want to reword it? Changing the tag requires that the correct tag be added, then the wrong tag deleted. This process would be easier if the tag could be edited directly.
